Out-of-character banter usually isn't supposed to affect the game, but sometimes a player's off-hand comment is so hilarious or appropriate that the DM sees fit to respond to it, either through modifiers, damage, or even roleplaying.

DM: There’s the upstairs living quarters which Princess Celestia has arranged for you to stay in.

Applejack: Speaking of said party… Are the rest of us all there too?
DM: Oh, yeah! Yeah. Of course you’re there. Right.
Applejack: That’s what I thought.
Pinkie Pie: The whole party’s here! Now we can have a PARTY PARTY!!
Rarity: …If this was my game, everyone would take 1d4 damage from that pun.
